Summary

Reliable partitioning of a cell suspension for single cell analysis is possible with the BD Rhapsody System. The suspension is distributed in a microwell cartridge in such a way that up to 25,000 cells are assigned individually to separate wells. The RNA of the individual cells is then bound to beads that can be individually identified by barcodes. Subsequently, sequencing libraries for targeted transcriptome panels or whole transcriptome analyzes are generated, which are then sequenced and evaluated bioinformatically.
In this system, it is also possible to simultaneously mark cells in advance with a large number of antibodies against surface structures, which can then also be quantified at the individual cell level. This information can then be bioinformatically combined with the transcriptome data.
In addition, multiplexing of several samples in one run is easily possible. This enables samples to be pooled in an analysis, whereby the individual cells and their information can be bioinformatically assigned to their sample origin, thus enabling direct comparability.
A continuous in-process control of the entire workflow guarantees a high level of quality assurance.
